Parkinson's Disease in Belgium

3. Oxidative Stress and α-Synuclein Aggregation
Industrial fructose massively generates ROS. Oxidative stress directly promotes misfolding and aggregation of α-synuclein — Lewy bodies (a pathological marker of Parkinson's). The substantia nigra is particularly vulnerable to oxidative stress (low glutathione content, high mitochondrial density).

- 6-Gingerol → dopaminergic protection: studies on MPTP models (animal Parkinson's model) — 40-60% reduction in dopaminergic neuronal loss with preventive gingerol
- NF-κB → neuroinflammation ↓: reduction of microglial activation and production of TNF-α and IL-1β in the substantia nigra
- Nrf2 activation: increases glutathione in the substantia nigra, protecting dopaminergic neurons against oxidative stress
- Autophagy: gingerol activates autophagy via AMPK → elimination of misfolded α-synucleins before aggregation

Does curcumin cross the blood-brain barrier?
Partially and in small quantities (lipophilic). Nanoparticle or liposomal forms have better brain penetration. Dietary curcumin from INTI primarily exerts its effects by systemically reducing inflammation.
Does INTI interact with levodopa (Parkinson's treatment)?
At normal dietary doses, no clinically significant interaction is documented. Curcumin can slightly inhibit CYP3A4 enzymes at very high doses — consult your neurologist for therapeutic doses.
