IL-6: Interleukin-6

IL-6 (Interleukin-6) is a pleiotropic cytokine, central to the acute phase response (CRP, fibrinogen, ferritin). It is a major player in polymyalgia rheumatica (PMR), giant cell arteritis (Horton's disease), severe COVID, and numerous inflammatory conditions. It is the therapeutic target of tocilizumab (anti-IL-6R).

  • Production: macrophages, T cells, fibroblasts, adipocytes
  • Signaling pathway: JAK1/JAK2 → STAT3

Biological Role

  • Acute phase response: induces CRP, fibrinogen, hepcidin, ferritin at the hepatic level
  • Differentiation of Th17 → maintenance of autoimmune diseases
  • Osteoclast activation → inflammatory osteoporosis
  • Chronic inflammation → cachexia, sarcopenia
