Curcumin: The Molecule from Turmeric
Curcumin (1,7-bis(4-hydroxy-3-methoxyphenyl)-1,6-heptadiene-3,5-dione) is the main bioactive orange-yellow polyphenol found in Curcuma longa (turmeric). It is a powerful inhibitor of NF-kB and IKK. It is considered one of the most studied natural anti-inflammatory compounds.

Mechanisms of Action
- IKK Inhibition → upstream blockade of NF-kB activation

The Problem of Bioavailability
Curcumin alone has extremely low oral bioavailability (~1%). Reasons:
- Low water solubility
- Rapid hepatic metabolism (glucuronidation)
- Massive biliary elimination
- Very short plasma half-life
Without a co-factor (piperine, lipids, special formulations), orally consumed curcumin has limited clinical effect. This is why the combination of curcumin + piperine is essential.

Conclusion: 20 mg of piperine combined with 2g of curcumin multiplies the plasma concentration of curcumin in humans by 20 (2000%). This is the foundational study that justifies the combination of turmeric + black pepper.
