TNF-α (Tumor Necrosis Factor alpha): The Master Cytokine of Inflammation

TNF-α: Tumor Necrosis Factor alpha

TNF-α (Tumor Necrosis Factor alpha) is a major pro-inflammatory cytokine produced primarily by activated macrophages. It plays a central role in rheumatoid arthritis, Crohn's disease, ankylosing spondylitis, and most chronic inflammatory diseases. It is the target of anti-TNF biotherapies (infliximab, adalimumab, etc.) — one of the most prescribed therapeutic classes in rheumatology/gastroenterology.

Molecular Characteristics

  • Type 2 cytokine (transmembrane then cleaved by TACE/ADAM17)
  • Receptors: TNFR1 (constitutive) and TNFR2 (inducible)
  • Serum half-life: 5-15 minutes
  • Production: macrophages, T lymphocytes, NK cells, mast cells, fibroblasts

Biological Effects

  • Activates NF-kB by binding to TNFR1 → amplified inflammatory cascade
  • Induces apoptosis of target cells via caspase-8
  • Stimulates the expression of COX-2, iNOS, MMPs
  • Recruits neutrophils and monocytes into tissues
  • Activates osteoclasts → bone resorption

Pathologies Involving Excess TNF-α

  • Rheumatoid Arthritis (RA)
  • Crohn's Disease and UC
  • Ankylosing Spondylitis
  • Psoriasis and Psoriatic Arthritis
  • Sepsis (septic shock)
  • Cancer Cachexia
  • Insulin Resistance (adipocyte TNF-α)

Documented Natural Inhibitors

  • [6]-gingerol: ↓ TNF-α by 30-50% in activated macrophages
  • Curcumin: inhibits TNF-α production via NF-kB and p38-MAPK
  • Piperine: potentiates curcumin's action on TNF-α
  • Resveratrol, EGCG (green tea), omega-3

TNF-α and INTI

INTI's combination of ginger + turmeric + piperine is designed to inhibit TNF-α through several complementary pathways. Documented systemic anti-inflammatory effect.
