3. NF-κB Inflammation and Tumor Microenvironment
Chronic sugar activates NF-κB → COX-2 → PGE2. PGE2 (prostaglandin E2) directly activates the aromatase gene promoter in mammary adipose tissue, creating a local inflammatory pro-tumor loop. This is the mechanism by which obesity specifically amplifies the risk of postmenopausal ER+ breast cancer.
4. Glycation and AGEs
Chronic sugar generates AGEs (Advanced Glycation End-products) which bind to RAGE receptors, activating NF-κB and PI3K in breast cancer cells. AGEs are markers of "cumulative sugar load" and are associated with a poorer prognosis in diagnosed breast cancers.
